    מחקר

    המעבדה  עוסקת בחקר מולקולארי ותאי של מחלות  סרטניות ודלקתיות.  במסגרת זו נחקרים במעבדה הנושאים הבאים:

    1. המאמץ בחקר מחלות סרטניות מתמקד הן ברובד הבסיסי והן בתחום הביוטכנולוגי יישומי. בתחום הבסיסי אנו חוקרים את מנגנון ייצור האנרגיה בתא הסרטני ובמאפיינים המבדילים אותו מתא נורמלי. מאמץ זה הביא לזיהוי חלבון (FerT ) המשתתף במערך ייצור האנרגיה של התא הסרטני אולם אינו קיים בזה של תאים נורמליים. מקורו של חלבון זה בתאי זרע שם הוא מאפשר ייצור יעיל של אנרגיה. נראה כי תאי הסרטן "אמצו" כלי יעיל זה אשר לא נמצא בתאים בריאים (להוציא תאי זרע) כדי ליעל ייצור האנרגיה בהם. אנו לומדים עתה באלו מנגנונים תורם האנזיםFerT    ליצור האנרגיה בתא הסרטני. במקביל פיתחנו מעכב סינתטי קטן מולקולארי אשר פוגע באופן ממוקד בפעילותו שלFerT  . חומר זה המכונה "אמיציל" גורם למותם של תאים סרטניים ביניהם גם תאים "אלימים" אשר הפכו לגרורות. זאת בלא שחומר זה ממית תאים נורמליים. "אמיציל" נמצא עתה בניסויי חיות כדי לבחון התאמתו לתרופה חדשה כנגד מחלות סרטניות. אנו נמצאים בתהליך של פיענוח המנגנונים התאיים המושפעים על יד אמיציל ומה מבדיל אותם מנתיבים מקבילים בתאים נורמליים.
    2. פרויקט נוסף במעבדה עוסק בחקר במחלות אוטו-אימוניות כרוניות. אנו מתמקדים בשלב זה בלימוד מנגנונים הקשורים להתפתחות לדלקות מעיים כרוניות:Colitis   ו-Crohn's . בשימוש בשיטות של מניפולציות גנטיות עלה בידינו לייצר במעבדה עכבר עמיד להתפתחותColitis . כלי זה אפשר לנו לזהות גן מפתח המקודד לחלבון-TMF/ARA160  והמבקר תהליכים דלקתיים שכיחים במעי אשר עשויים להתפתח לגידולים סרטניים. אנו חוקרים עתה כיצד משפיע TMF/ARA160 על רגישות יונקים לדלקות כגון Colitis   ו-Crohn's .
    3.  נושא נוסף הנחקר במעבדה קשור בעמידות תאי מח עצביים לתנאי עקה. לנושא חשיבות רבה שכן נודעות לו השלכות על הבנת הגורמים המולקולאריים התורמים לפגיעה בתאי עצב תחת תנאי עקה כגון חוסר בחומרי מזון וחמצן בעקבות אירוע מוחי. בשימוש במבחנים שונים זיהינו חלבון המכונהTRNP  המסייע לתאי עצב לשרוד מצב של העדר חומרי מזון. חלבון זה מצוי בגרעין תאי עצב והוא מפעיל מערך ביטוי גנים כתגובה לתנאי עקה. אנו לומדים עתה מהן הרשתות המולקולאריות המופעלות על ידי מבקר שעתוק זה. תוצאות המחקר צפויות להעמיק הבנתינו במנגנונים אשר יאפשרו מזעור נזקים לתאי עצב אשר נחשפו לתנאי עקה שונים.

    לסיכום ניתן לומר כי הפרויקטים השונים במעבדה מחברים בין מחקר בסיס לבין מחקר ביו-רפואי בעל השלכות של התמודדות עם מחלות שונות באדם.
